Fluorescence microscopy is the primary tool for studying complex processes inside individual living cells. Technical advances in both molecular biology and microscopy have made it possible to image cells from many genetic and environmental backgrounds. These images contain a vast amount of information, which is often hidden behind various sources of noise, convoluted with other information and stochastic in nature. Accessing the desired biological information therefore requires new tools of computational image analysis and modeling. Here, we review some of the (...) recent advances in computational analysis of images obtained from fluorescence microscopy, focusing on bacterial systems. We emphasize techniques that are readily available to molecular and cell biologists but also point out examples where problem‐specific image analyses are necessary. Thus, image analysis is not only a toolkit to be applied to new images but also an integral part of the design and implementation of a microscopy experiment. (shrink)

Understanding is a valued trait in any epistemic practice, scientific or not. Yet, when it comes to characterizing its nature, the notion has not received the philosophical attention it deserves. We have set ourselves three tasks in this paper. First, we defend the importance of this endeavor. Second, we consider and criticize a number of proposals to this effect. Third, we defend an alternative account, focusing on abilities as the proper mark of understanding.

Reuben Hersh confided to us that, about forty years ago, the late Paul Cohen predicted to him that at some unspecified point in the future, mathematicians would be replaced by computers. Rather than focus on computers replacing mathematicians, however, our aim is to consider the (im)possibility of human mathematicians being joined by “artificial mathematicians” in the proving practice—not just as a method of inquiry but as a fellow inquirer.

From 2009 to 2013 the workplace innovation project “My Enterprise 2.0” was carried out in the region of Utrecht in the Netherlands in order to strengthen the workplace innovation capability of small and medium-sized enterprises. Participating enterprises completed a questionnaire regarding the “workplace innovativeness” of their company. A workplace innovation intervention was then implemented by some of the companies, while other companies chose not to take part. At the end of the project, a second questionnaire indicated that those companies that (...) implemented interventions had a significantly higher score with regard to overall workplace innovativeness. The companies without such interventions reported a small decrease. While the companies in the region had higher workplace innovativeness scores relative to a national reference group both before and after the project, the increase in the “workplace innovativeness” of the regional SMEs that experienced interventions suggests that the project proved beneficial to their continued “workplace innovativeness.” Moreover, these companies also reported positive effects on company performance, achieving company goals and improving labor productivity. While self?evaluation leads to valuable results in some schools, it appears that in other schools this is true only to a lesser extent or not at all. This raises the question of how differences in the results of self?evaluations can be explained. This study looks at to what extent the results of self?evaluation are determined by the way in which self?evaluation is conducted, by characteristics relating to the general functioning of the school and by the support which schools enjoy. One (...) thousand seven hundred and eighty?six school principals and team members from 96 schools (primary and secondary) were surveyed by means of a written questionnaire. The data collected were then analysed using multi?level structural equation modelling. The results provide strong empirical evidence that ?attitude with regard to self?evaluation?, ?self?evaluation as a policy action? and ?self?evaluation as an act of research? are powerful predictors of the quality of self?evaluations. Despite the proliferation of research on ethical leadership, there remains a limited understanding of how specifically the assumingly moral component of this leadership style affects employee behavior. Taking an identity perspective, we integrate the ethical leadership literature with research on the dynamics of the moral self-concept to posit that ethical leadership will foster a sense of moral identity among employees, which then inspires followers to adopt more ethical actions, such as increased organization citizenship behavior. We further argue that these identity (...) effects should be more pronounced when leaders are perceived to be group prototypical, as their actions then speak louder to followers’ sense of identity. Two studies—a scenario experiment with 138 participants and a field study with 225 employees—provided support for our hypothesized moderated mediation model. Perceived ethical leadership positively affected OCB via followers’ moral identity but only under conditions of high perceived leader group prototypicality. We discuss how the identity pathway of ethical leadership can facilitate novel theorizing about moral transference. Our findings also suggest that, when hiring external ethical leaders or training internal managers, practitioners are well advised to consider that these individuals may only be effective in morally transforming followers when they are perceived as prototypical for the group. Wilson & Sober's discussion of group selection is marred by the absence of plausible examples of human group-level behavioral adaptation. The trait of authoritarianism is one possible example of such an adaptation. It reduces within-group variance in reproductive success, manifests itself more strongly in response to group-level threat, and is found in a variety of cultures.

We investigated, by means of the Reverse Correlation Task, visual representations of the culturally dominating group of local people held by sojourners as a function of their degree of cross-cultural adaptation. In three studies, using three different methods with three independent samples of sojourners and seven independent samples of Portuguese and US-American raters, we gathered clear evidence that poor adaptation goes along with more negative representations of locals. This indicates that sojourner adaptation is reflected, at a social-cognitive level, in the (...) valence of outgroup representations. (shrink)

This study explored LaRose and Eastin's model of media attendance, within a European context. It extended the uses and gratifications paradigm within the framework of social cognitive theory by instituting new operational measures of gratifications sought, reconstructed as outcome expectations. Although the model of media attendance offers some promising steps forward in measuring media selectivity and usage, and to some extent is applicable to another context of media use, the relative importance of outcome expectancies in explaining media usage and selectivity (...) is not fully supported. This book, formed as a series of essays in honour of Professor Carl Baudenbacher, addresses the very art of judicial reasoning, and features contributions from many of the foremost current or former national, supranational, or international judges. Cyclic adenosine monophosphate (cAMP) and cAMP‐dependent protein kinase A (PKA) are evolutionary conserved molecules with a well‐established position in the complex network of signal transduction pathways. cAMP/PKA‐mediated signaling pathways are implicated in many biological processes that cooperate in organ development including the motility, survival, proliferation and differentiation of epithelial cells. Cell surface polarity, here defined as the anisotropic organisation of cellular membranes, is a critical parameter for most of these processes. Changes in the activity of cAMP/PKA elicit a variety of (...) effects on intracellular membrane dynamics, including membrane sorting and trafficking. One of the most intriguing aspects of cAMP/PKA signaling is its evolutionary conserved abundance on the one hand and its precise spatial–temporal actions on the other. Taking as its point of departure the works of Jacques Ellul, Sven Birkerts, George Steiner, Uwe Poerksen, and others, this article explores the status of language in a technicized civilization. It is argued that language has devolved under the impact of technology, particularly in the dimension of values and ethics. This diagnosis points to the way from which a possible cure may emerge.

How do we ascribe subjective probability? In decision theory, this question is often addressed by representation theorems, going back to Ramsey (1926), which tell us how to define or measure subjective probability by observable preferences. However, standard representation theorems make strong rationality assumptions, in particular expected utility maximization. How do we ascribe subjective probability to agents which do not satisfy these strong rationality assumptions? This paper attempts to answer the question of what defines mnemonic confabulation vis-à-vis genuine memory. The two extant accounts of mnemonic confabulation as “false memory” and as ill-grounded memory are shown to be problematic, for they cannot account for the possibility of veridical confabulation, ill-grounded memory, and wellgrounded confabulation. This paper argues that the defining characteristic of mnemonic confabulation is that it lacks the appropriate causal history. In the confabulation case, there is no proper counterfactual dependence of the state of (...) seeming to remember on the corresponding past representation. (shrink)

The global method safety account of knowledge states that an agent’s true belief that p is safe and qualifies as knowledge if and only if it is formed by method M, such that her beliefs in p and her beliefs in relevantly similar propositions formed by M in all nearby worlds are true. This paper argues that global method safety is too restrictive. First, the agent may not know relevantly similar propositions via M because the belief that p is the (...) only possible outcome of M. Second, there are cases where there is a fine-grained belief that is unsafe and a relevantly similar coarse-grained belief that is safe and where both beliefs are based on the same method M. Third, the reliability of conditional reasoning, a basic belief-forming method, seems to be sensitive to fine-grained contents, as suggested by the wide variation in success rates for thematic versions of the Wason selection task. According to T. M. Scanlon's 'buck-passing' analysis of value, x is good means that x has properties that provide reasons to take up positive attitudes vis-à-vis x. Some authors have claimed that this idea can be traced back to Franz Brentano, who said in 1889 that the judgement that x is good is the judgement that a positive attitude to x is correct ('richtig'). The most discussed problem in the recent literature on buckpassing is known as the 'wrong kind of (...) reason' problem (the WKR problem): it seems quite possible that there is sometimes reason to favour an object although that object is not good and possibly very evil. The problem is to delineate exactly what distinguishes reasons of the right kind from reasons of the wrong kind. In this paper we offer a Brentano-style solution. We also note that one version of the WKR problem was put forward by G. E. Moore in his review of the English translation of Brentano's Vom Ursprung sittlicher Erkenntnis. Before getting to how our Brentano-style approach might offer a way out for Brentano and the buck-passers, we briefly consider and reject an interesting attempt to solve the WKR problem recently proposed by John Skorupski. Sven Bernecker presents an analysis of the concept of propositional (or factual) memory, and examines a number of metaphysical and epistemological issues crucial to the understanding of memory. -/- Bernecker argues that memory, unlike knowledge, implies neither belief nor justification. There are instances where memory, though hitting the mark of truth, succeeds in an epistemically defective way. This book shows that, contrary to received wisdom in epistemology, memory not only preserves epistemic features generated by other epistemic sources but also (...) functions as a source of justification and knowledge. According to the causal theory of memory argued for in this book, the dependence of memory states on past representations supports counterfactuals of the form: if the subject hadn't represented a given proposition in the past he wouldn't represent it in the present. The book argues for a version of content externalism whereupon the individuation of memory contents depends on relations the subject bears to his past physical or social environment. Moreover, Bernecker shows that memory doesn't require identity, but only similarity, of past and present attitudes and contents. The notion of content similarity is explicated in terms of the entailment relation. (shrink)

Is more information always better? Or are there some situations in which more information can make us worse off? Good (1967) argues that expected utility maximizers should always accept more information if the information is cost-free and relevant. But Good's argument presupposes that you are certain you will update by conditionalization. If we relax this assumption and allow agents to be uncertain about updating, these agents can be rationally required to reject free and relevant information. Since there are good reasons (...) to be uncertain about updating, rationality can require you to prefer ignorance. (shrink)

Justification as Ignorance offers an original account of epistemic justification as both non-factive and luminous, vindicating core internalist intuitions without construing justification as an internal condition knowable by reflection alone. Sven Rosenkranz conceives of justification, in its doxastic and propositional varieties, as a kind of epistemic possibility of knowing and of being in a position to know. His account contrasts with recent alternative views that characterize justification in terms of the metaphysical possibility of knowing. Instead, he develops a suitable (...) non-normal multi-modal epistemic logic for knowledge and being in a position to know that respects the finding that these notions create hyperintensional contexts. He also defends his conception of justification against well-known anti-luminosity arguments, shows that the account allows for fruitful applications and principled solutions to the lottery and preface paradoxes, and provides a metaphysics of justification and its varying degrees of strength that is compatible with core assumptions of the knowledge-first approach and disjunctivist conceptions of mental states. This paper provides a novel argument for granting memory the status of a generative source of justification and knowledge. Memory can produce justified output beliefs and knowledge on the basis of unjustified input beliefs alone. The key to understanding how memory can generate justification and knowledge, memory generativism, is to bear in mind that memory frequently omits part of the stored information. The proposed argument depends on a broadly reliabilist approach to justification.

This book investigates central issues in the philosophy of memory. Does remembering require a causal process connecting the past representation to its subsequent recall and, if so, what is the nature of the causal process? Of what kind are the primary intentional objects of memory states? How do we know that our memory experiences portray things the way they happened in the past? Given that our memory is not only a passive device for reproducing thoughts but also an active device (...) for processing stored thoughts, when are thoughts sufficiently similar to be memory-related? The Metaphysics of Memory defends a version of the causal theory of memory, argues for direct realism about memory, proposes an externalist response to skepticism about memory knowledge, and develops a contextualist account of the factivity constraint on memory. In the first part of this paper, I clear the ground from frequent misconceptions of the relationship between fact and value by examining some uses of the adjective “natural” in ethical controversies. Such uses bear evidence to our “natural” tendency to regard nature as the source of ethical norms. I then try to account for the origins of this tendency by offering three related explanations, the most important of which is evolutionistic: if any behaviour that favours our equilibrium with the (...) environment is potentially adaptive, nothing can be more effective for this goal than developing an attitude toward the natural world that considers it as a dispenser of sacred norms that must be invariably respected. By referring to the Aristotelian notion of human flourishing illustrated in the first part of the paper, in the second I discuss as a case study some ethical problems raised by mini-chips implantable in our bodies. I conclude by defending their potential beneficial effects of such new technological instruments. (shrink)

This paper challenges the standard conception of memory as a form of knowledge. Unlike knowledge, memory implies neither belief nor justification.

It is widely recognized that lives and activities can be meaningful or meaningless, but few have appreciated that they can also be anti-meaningful. Anti-meaning is the polar opposite of meaning. Our purpose in this essay is to examine the nature and importance of this new and unfamiliar topic. In the first part, we sketch four theories of anti-meaning that correspond to leading theories of meaning. In the second part, we argue that anti-meaning has significance not only for our attempts to (...) theorize about meaning in life, but also for our ability to lead meaningful lives in the modern world. Abstract“Gamification” refers to adding game-like elements to non-game activities so as to encourage participation. Gamification is used in various contexts: apps on phones motivating people to exercise, employers trying to encourage their employees to work harder, social media companies trying to stimulate user engagement, and so on and so forth. Here, I focus on gamification with this property: the game-designer (a company or other organization) creates a “game” in order to encourage the players (the users) to bring about certain outcomes (...) as a side effect of playing the game. The side effect might be good for the user (e.g., improving her health) and/or good for the company or organization behind the game (e.g., advertising their products, increasing their profits, etc.). The “players” of the game may or may not be aware of creating these side effects; and they may or may not approve of/endorse the creation of those side effects. The organizations behind the games, in contrast, are typically directly aiming to create games that have the side effects in question. These aspects of gamification are puzzling and interesting from the point of view of philosophical analyses of agency and responsibility for outcomes. In this paper, I relate these just-mentioned aspects of gamification to some philosophical discussions of responsibility gaps, the ethics of side effects (including the Knobe effect and the doctrine of double effect), and ideas about the relations among different parties’ agency.
